Yes, when the timing mark is on "0" that is Top Dead Centre (TDC) and the valves are fully closed on the cylinder that is firing (which will be either No: 1 or No: 4 cylinder).
When you do the distributor timing, it is set at about 10 degrees Before Top Dead Centre "BTDC". This allows time for the burn to expand and be compressed before sending the piston back down the bore.
If you are setting the tappet adjustment you should be able to put the timing mark at TDC and adjust four of the tappets then rotate the crank once back to TDC and adjust the other four.
